Vector: pGem T easy vector
Resistances: Ampicillin
Bacterial Strain: DH5a
Insert: E. coli beta galactosidase (lacZ) gene; 670 bp fragment amplified by using two internal primers
Description of cloning procedure:
Amplified by PCR with LacZ F and LacZ R primers on the E.coli LacZ gene and cloned in pGEM T easy vector
Intended Use: It has been used to obtain both sense and anti-sense DIG-labeled "LacZ riboprobes"
Special Remarks: To obtain the antisense riboprobe, you can linearize with the Sal1 restriction enzyme and retro-transcribe by using T7 RNA polymerase; for the sense riboprobe, useful as negative control, digest with the NcoI RE and retrotranscribe with the SP6 RNA pol.
