Model System - EPN
Human non-transformed and differentiated epithelial prostate cell line
Figure legend:
Description
EPN cells in culture, as prostate epithelial cells in vivo, express cytokeratin 5 and 14 whereas they do not express desmin. The epithelial origin of EPN cells is strengthened by the observation that EPN cells express an active form of the cytoplasm tyrosine kinase Pyk2. In fact, Pyk2 is expressed in the normal human prostate epithelia but not in the stromal component of the prostate (Stanzione et al., 2001).
Finally, clonal production of the prostate-specific antigen (PSA) could be detected in EPN cells.
A nontransformed differentiated prostate epithelial cell in vitro, like the EPN cells, will enrich the scenario of prostate model systems in vitro and will prove useful in a wide range of studies in cellular genetics, gene regulation, pharmacology, and aging.
Culture conditions: Keratinocyte-serum free medium (SFM) ( Gibco-Brl Milan Italy)supplemented with bovine pituitary excrat (10mg/ml), epidermal growth factor EGF (10ng/ml),cholera toxin (10ng/ml), 5% foetal calf serum (FCS) (Gibco-Brl, Life technologies, Paisley Sctland UK).
